Target Information
The Pax gene family encodes transcription factors active in specific tissues during early embryonic development. Pax proteins contain a DNA-binding domain encoded by the paired box, enabling them to regulate genetic programs. Mutations in Pax genes are associated with developmental disorders such as:
- Pax-3: Waardenburg syndrome
- Pax-6: Aniridia
- Pax-2: Wilms tumor
During embryogenesis, Pax-2 is expressed in the developing kidney, particularly in condensing metanephric mesenchyme and early epithelial structures. Pax-2 expression decreases as tubular epithelium matures but persists in Wilms tumor, correlating with poorly differentiated epithelial cell proliferation. Expression of the Wilms tumor suppressor WT1 coincides with Pax-2 down-regulation, suggesting WT1 directly represses Pax-2 transcription.
