Target Information
The E3 ubiquitin-protein ligase NEDD4 is a Ubl conjugation pathway protein containing one HECT (E6AP-type E3 ubiquitin-protein ligase) domain and four WW domains. It participates in the degradation pathway of VEGFR-2/KDFR, independently of its ubiquitin-ligase activity.
NEDD4 acts as a potential proto-oncogene that negatively regulates PTEN via ubiquitination, similar to the Mdm2–p53 paradigm. It is also involved in viral budding and regulates epithelial Na⁺ channels, contributing to arterial hypertension.
NEDD4 protein localizes to the sarcolemmal region of muscle fibers, and increased expression is associated with unloaded or denervated muscles and decreased Notch1 expression, acting as a down-regulator of Notch-1 gene expression.
