Target Information
IGF1R (Insulin-like Growth Factor 1 Receptor, CD221) is a receptor tyrosine kinase activated by IGF-1 and IGF-2, implicated in various cancers. The protein is a tetramer consisting of two alpha and two beta subunits. The alpha subunits bind IGF-1 with higher affinity than IGF-2, leading to conformational change and autophosphorylation at positions 1134, 1135, and 1136. Activated IGF-1R phosphorylates substrate proteins, sequentially activating RAS, RAF, and MAPK isoforms (ERK, p38, JNK), which drive gene transcription for proliferation.
