Recommended positive control tissue: Prostate carcinoma, though other tissues may also be suitable.

The antibody is intended for professional laboratory use in detecting ERG protein in formalin-fixed, paraffin-embedded tissue via manual qualitative IHC testing. It should be used after primary tumor diagnosis by conventional histopathology.

ERG (ETS-related gene) is a proto-oncogene encoding a nuclear transcription factor involved in hematopoietic and endothelial development. It is continuously expressed in endothelial cells, blood and lymphatic vessels, and bone marrow stem cells. ERG expression is seen in endothelial neoplasms such as hemangioendothelioma, angiosarcoma, and Kaposi sarcoma. It is overexpressed due to gene rearrangement in prostate adenocarcinoma and various other tumors.

ERG serves as a sensitive and specific marker for endothelial differentiation, offering easier interpretation due to its nuclear localization, which permits double staining with cytoplasmic markers like podoplanin. Among carcinomas, ERG is highly specific for prostate tissue.

Pretreatment of deparaffinized tissue with heat-induced or enzymatic epitope retrieval is recommended. Standard IHC staining involves sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate, followed by visualization under a light microscope.

Positive tissue controls must accompany each staining procedure. These should include both positive and negative staining components. External control tissues should be processed similarly to patient samples and selected for low-level positive staining to ensure assay sensitivity and reagent stability. Negative controls should confirm absence of non-specific staining.
