The G-protein-coupled receptor (GPCR) superfamily includes approximately 600–1,000 members, representing the largest known class of molecular targets with therapeutic significance.
GPR31 is a high-affinity receptor for 12-(S)-hydroxy-5,8,10,14-eicosatetraenoic acid (12-S-HETE), an arachidonic acid metabolite secreted by platelets and tumor cells.
12-(S)-HETE induces endothelial cell retraction, facilitating invasive cell access to the subendothelial matrix — a key step in extravasation and metastasis.
Ligand binding activates MAPK and NF-κB signaling pathways.
