Infection with human cytomegalovirus (HCMV) causes relocation to Golgi apparatus and cytoplasmic vacuoles containing HCMV proteins (glycoprotein B, pp28). Interaction with HCMV/HHV-5 protein vMIA/UL37 results in relocalization from ER to mitochondria.

Background

  • Cofactor: Binds one 4Fe-4S cluster coordinated with three cysteines and an exchangeable S-adenosyl-L-methionine.
  • Function: Involved in antiviral defense. May impair virus budding by disrupting lipid rafts at the plasma membrane, essential for budding of many viruses. Acts by binding and inactivating FPPS, an enzyme involved in cholesterol, farnesylated and geranylated proteins, ubiquinones, dolichol, and heme synthesis. Plays a major role in the antiviral state induced by type I and II interferons. Displays antiviral effects against HIV-1, hepatitis C virus, human cytomegalovirus, and alphaviruses, but not vesiculovirus.
  • Induction: Induced by interferon type I, type II, and LPS. Little or no induction by interferon gamma in monocytic cell lines. Upregulated by infection with HCMV, hepatitis C virus, yellow fever virus, and Sendai virus via type I interferon pathway.
  • Miscellaneous: Up-regulated in atherosclerosis. Latent viruses like HCMV may contribute to atherogenesis by initiating local inflammation, inducing RSAD2 expression, modulating lipid synthesis, and contributing to lipid accumulation.
  • Similarity: Belongs to the RSAD2 family.
  • Subunit: Interacts with FPPS.
  • Subcellular Location: Associates with the cytosolic side of the endoplasmic reticulum; relocates to Golgi apparatus and vacuoles containing viral proteins upon infection.
