Background
The protein encoded by this gene is a cytokine belonging to the tumor necrosis factor (TNF) ligand family. It preferentially induces apoptosis in transformed and tumor cells without affecting most normal cells, despite being expressed in many normal tissues. This protein binds to several TNF receptor superfamily members including TNFRSF10A/TRAILR1, TNFRSF10B/TRAILR2, TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4, and possibly TNFRSF11B/OPG. Its activity can be modulated by decoy receptors that cannot induce apoptosis. Binding of TRAIL to its receptors triggers activation of MAPK8/JNK, caspase 8, and caspase 3. Alternatively spliced transcript variants encoding different isoforms have been identified for this gene.
