Background

The protein encoded by this gene belongs to the SMAD family, homologous to the Drosophila gene mothers against decapentaplegic (Mad) and the C. elegans gene Sma. SMAD proteins act as signal transducers and transcriptional modulators mediating multiple signaling pathways. Smad1 mediates bone morphogenetic protein (BMP) signals involved in cell growth, apoptosis, morphogenesis, development, and immune responses. Upon BMP stimulation, Smad1 is phosphorylated and activated by BMP receptor kinases, forming a complex with SMAD4 for transcriptional regulation. It is targeted by SMAD-specific E3 ubiquitin ligases such as SMURF1 and SMURF2, undergoing ubiquitination and proteasomal degradation.
