Target Information
MAP4K3 (also known as GLK) is a member of the STE20 family of serine/threonine protein kinases. It contains an N-terminal catalytic domain and a C-terminal regulatory domain. MAP4K3 acts upstream of MEKK1 and is stimulated by UV radiation and TNF-alpha, leading to activation of the JNK signaling pathway. It can directly activate PKC-θ during TCR signaling. Enhanced MAP4K3 expression has been observed in systemic lupus erythematosus, contributing to activation of PKC-θ and IKK in T cells. MAP4K3 also functions as a pro-apoptotic kinase, orchestrating BAX activation via modulation of PUMA, BAD, and BIM. The kinase activity is stimulated by UV radiation and TNF-alpha and specifically activates the c-Jun N-terminal kinase (JNK) pathway. MAP4K3 was previously referred to as RAB8-interacting protein-like 1 (RAB8IPL1) and is now known as mitogen-activated protein kinase kinase kinase 3 (MAP4K3).
