Target Information
GAD-65와 GAD-67은 group II decarboxylase family에 속하는 효소로, L-glutamic acid로부터 GABA(gamma-aminobutyric acid)를 생성하는 과정의 속도 제한 단계(rate-limiting step)를 촉매합니다.
두 GAD 단백질 모두 뇌에서 발견되지만, GAD-65는 신경 말단의 시냅스 소포막에 주로 존재하며, GAD-67은 세포 전체에 분포합니다.
GAD-67은 기본적인 GABA 합성을 담당하며, 신경전달 과정에서 GABA 요구가 증가할 경우 GAD-65가 일시적으로 활성화되어 GABA 생산을 돕습니다.
GAD-65의 결핍은 GABA 신경전달을 저해할 수 있으며, GAD-67의 결핍은 치명적입니다.
대체 스플라이싱으로 인해 GAD-67에는 두 가지 isoform(GAD-67 및 GAD-25)이 존재합니다.
