Target Information

The second messengers cAMP and cGMP are key regulatory molecules involved in a wide variety of signal transduction pathways. Their levels are controlled by synthesis through nucleotide cyclases and hydrolysis by cyclic nucleotide phosphodiesterases (PDEs). PDEs form a superfamily of enzymes that convert cyclic nucleotides to their corresponding nucleoside monophosphates.
PDE6 acts as the effector enzyme in the G protein-mediated signal transduction cascade in the visual system. It consists of multiple subunits: rod and cone-specific catalytic subunits (alpha’, alpha, and beta), an inhibitory gamma subunit, and a delta subunit of unknown function. The catalytic core includes alpha’/alpha’ homodimers in cones and alpha/beta heterodimers in rods. The C-terminus of both catalytic and inhibitory subunits undergoes methylation, myristoylation, and prenylation, essential for proper complex assembly and membrane association.
