Target Information
TNFR1 (Tumor necrosis factor receptor 1)는 TNF 수용체 초가족에 속하며, TNF-α의 주요 수용체 중 하나입니다. TNFR1은 사이토카인 매개 신호전달, NF-κB 경로의 양성 조절, 혈관신생의 양성 조절, 유전자 발현의 음성 조절 등 다양한 생물학적 과정에 관여합니다.
세포 외 도메인은 순환계로 방출되어 용해형 TNFR1(sTNFR1)을 형성합니다. 인간에서는 TNFR1 유전자가 염색체 12번에 위치합니다.
BCL2-associated athanogene 4 (BAG4/SODD), TRADD, TRAF2 단백질이 TNFR1과 상호작용하여 수용체 매개 신호전달을 조절합니다. TNFR1의 세포외 도메인에 존재하는 생식세포 돌연변이는 상염색체 우성 주기성 발열 증후군과 관련이 있으며, 수용체 제거 장애가 질병의 기전으로 제시됩니다.
