Target Information
VPRBP acts both as a substrate recognition component of E3 ubiquitin-protein ligase complexes and as an atypical serine/threonine-protein kinase. It plays key roles in processes such as cell cycle regulation, telomerase activity, and histone modification.
It functions as a substrate-specific adapter in the CUL4A-RBX1-DDB1-DCAF1/VPRBP complex, mediating ubiquitination and degradation of proteins such as NF2.
VPRBP also recognizes methylated proteins via its chromo domain, leading to their ubiquitination. It is involved in B-cell development through RAG1-mediated ubiquitination, limiting error-prone repair during V(D)J recombination.
Additionally, it participates in telomerase inhibition by mediating TERT degradation and acts as a serine/threonine-protein kinase that phosphorylates Thr-120 of histone H2A (H2AT120ph), repressing transcription of tumor suppressor genes.
During HIV infection, VPRBP is hijacked by viral proteins (Vpr or Vpx) to manipulate host cell cycle and enhance viral replication.
