Product Specific Information

The 5H10 monoclonal antibody recognizes the mouse CD8α chain. It has been used for flow cytometry and inhibition of T cell responses to IL-2. Each lot is tested using freshly harvested mouse splenocytes.

• When diluting, prepare only the amount needed for one week.
• Analyze stained cells within 18 hours for optimal results.
• For Pacific Orange™ conjugates: excitation 405 nm / emission 551 nm.
• Matching isotype control: Rat IgG2b Pacific Orange™.
• Recommended detection filter: 575 or 585 nm band pass.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein that plays a crucial role in T cell activation and differentiation. It forms heterodimers (CD8α/CD8β) acting as a co-receptor for MHC-I molecules, while CD4 serves as a co-receptor for MHC-II. CD8 is expressed on subsets of T cells, NK cells, monocytes, and dendritic cells.

Upon MHC-I/peptide complex ligation, CD8 recruits Lck kinase, initiating signaling that leads to cytokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are essential for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
