Target Information

NUDT16 is an RNA-binding and decapping enzyme that catalyzes cleavage of the cap structure of snoRNAs and mRNAs in a metal-dependent manner. It is part of the U8 snoRNP complex required for the accumulation of mature 5.8S and 28S rRNA. The enzyme has diphosphatase activity, removing m7G and/or m227G caps from U8 snoRNA and leaving a 5′ monophosphate on the RNA. It also acts on mRNAs, but does not hydrolyze cap analog structures like m7GpppG. NUDT16 exhibits broad substrate specificity with manganese or cobalt as cofactors and binds U8 snoRNA independently of metal ions. It may regulate snoRNA and mRNA degradation and also acts as a phosphatase, hydrolyzing non-canonical purine nucleotides (IDP, dITP, GDP, etc.) to their monophosphate derivatives, thereby preventing incorporation of non-canonical purines into nucleic acids and avoiding chromosomal lesions.
