Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily. It was first identified as a surface antigen on Hodgkin’s and Reed-Sternberg cells using the monoclonal antibody Ki-1.
The ligand for CD30 is CD30L (CD153), and their interaction mediates various cellular processes including proliferation, activation, differentiation, and apoptosis.
CD30 plays a crucial role in the pathophysiology of Hodgkin’s disease and other CD30-positive lymphomas. It also functions as a costimulatory molecule during thymic negative selection.

CD30 expression is observed in:

  • Hodgkin’s and Reed-Sternberg cells
  • Certain non-Hodgkin’s lymphomas (e.g., Burkitt’s lymphoma)
  • Virus-infected T and B cells
  • Activated normal T and B cells

In T cells, CD30 is expressed on subsets that produce Th2-type cytokines and on CD4+/CD8+ thymocytes co-expressing CD45RO and IL4 receptor.
The soluble form (sCD30) serves as a marker of Th2 immune response.
CD30 interacts with TRAF2 and TRAF5, leading to NF-κB activation and promoting apoptosis regulation, limiting autoreactive CD8 effector T cell proliferation, and protecting against autoimmunity.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, and most Anaplastic Large Cell Lymphomas (ALCL), as well as in embryonal carcinomas.
