Product Specific Information

Description:
The eBioY1/82A antibody reacts with CD68, a member of the sialomucin family related to lysosomal-associated membrane proteins and scavenger receptors. CD68 is mainly intracellular, found in late endosomes, and expressed on myeloid-derived cells such as monocytes, macrophages, dendritic cells, neutrophils, and basophils. It may play roles in antigen processing/presentation and lysosomal membrane protection.

Staining with eBioY1/82A shows surface staining on B cells and minor monocyte populations, while intracellular staining detects about 50% of B cells and most monocytes.

Applications Reported:

  • Flow cytometric analysis of normal human peripheral blood cells.

Applications Tested:

  • Pre-titrated and tested for intracellular staining and flow cytometry at 5 µL (0.5 µg)/test.
  • Recommended cell count: 10⁵–10⁸ cells/test (empirically determined).

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD68 (Macrosialin):
A 110 kDa integral membrane glycoprotein predominantly expressed on intracellular lysosomes of monocytes and macrophages. Also found in dendritic cells and granulocytes. CD68 is implicated in phagocytic activity and lysosomal metabolism. Expression is observed in tonsil interdigitating reticulum cells, histiocytic lymphoma, AML, and granulocytic sarcoma. Elevated expression noted in CD34+ cells in various malignancies, including AML.
