Target Information
Nalcn is a voltage-independent, cation-nonselective channel permeable to Na⁺, K⁺, and Ca²⁺ ions. It regulates resting membrane potential and neuronal excitability. Neuropeptides such as neurotensin and substance P activate NALCN via SRC family kinase-dependent pathways. Nalcn activity is modulated by GPCRs, essential for respiratory rhythm, neonatal survival, and systemic osmoregulation. It contributes to intestinal pacemaking and neuronal excitability in substantia nigra pars reticulata.
Diseases associated with NALCN include congenital contractures of limbs and face, hypotonia, developmental delay, and infantile psychomotor retardation with characteristic facies.
