• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

Alpha 2-Macroglobulin (A2M) is an approximately 720-kDa homotetrameric glycoprotein composed of four identical ~180-kDa subunits.
A2M shares with other alpha-macroglobulins (e.g., complement components C3, C4, and pregnancy zone protein PZP) a broad ligand-binding capacity, serving as a humoral defense barrier against foreign peptides in plasma.
A2M interacts with and captures virtually any proteinase, functioning as a panprotease inhibitor.
In the brains of Alzheimer’s disease patients, A2M has been localized to diffuse amyloid plaques, supporting its role in AD pathophysiology.
