Target Information

Tau is a neuronal microtubule-associated protein found predominantly on axons. Its primary function is to promote tubulin polymerization and stabilize microtubules. The C-terminus binds axonal microtubules while the N-terminus interacts with neural plasma membrane components, acting as a linker between both structures.

Tau localization determines axonal polarity within neurons. Short isoforms contribute to cytoskeletal plasticity, while longer isoforms aid in stabilization. In its hyper-phosphorylated form, Tau is the major component of paired helical filaments (PHF), which form neurofibrillary lesions characteristic of Alzheimer’s disease (AD).

Hyper-phosphorylation impairs Tau’s microtubule-binding ability, leading to microtubule destabilization and neuronal degeneration in AD. Multiple kinases including GSK-3β, PKA, CDK5, and casein kinase II are known to phosphorylate Tau. Hyper-phosphorylated Tau is also found in other neurodegenerative disorders such as Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
