Target Information

IL-33R, also known as ST2, is a receptor for IL-33 and a member of the IL-1 receptor family.
Its extracellular domain contains three Ig-like domains that interact with the co-receptor IL-1RAcP and ligand IL-33.
Binding of IL-33 triggers association with adaptor proteins (MyD88, IRAK1, IRAK4, TRAF6), activating NF-κB and MAPK pathways and promoting cytokine release (IL-5, IL-6, IL-8, IL-13).
Two isoforms exist due to alternative splicing:

• ST2L: membrane-bound form
• ST2S: soluble form acting as a decoy to dampen IL-33 signaling

IL-33R is mainly expressed by mast cells, basophils, and eosinophils. In mice, expression is also observed in ILC2 and Th2 cells.
IL-33R signaling is implicated in immune conditions including allergies, arthritis, atherosclerosis, and sepsis.
