The GeneArt Gibson Assembly HiFi Cloning Kit enables seamless DNA assembly and cloning through a one-step, 15–60 minute isothermal reaction that links multiple overlapping DNA fragments. This approach allows uniform assembly of DNA fragments using complementary overlaps, suitable for both small and large constructs, including single or multiple inserts. The resulting products can be used for downstream applications such as transformation, PCR, and rolling circle amplification (RCA).
The kit includes master mix, positive control, water, and ElectroMAX DH10B electrocompetent E. coli.

Seamless Assembly of Multiple Fragments
The GeneArt Gibson Assembly HiFi cloning process uses a proprietary enzymatic mix to combine up to six fragments with shared terminal homology, leaving no extra sequences or scars. After 15–60 minutes of incubation, the reaction mixture can be transformed into competent cells, producing clones ready for analysis the next day. The required 20–40 bp homology can be designed during fragment synthesis or introduced via PCR amplification with custom oligos. This enzyme mix creates a seamless, covalently bound DNA construct for high efficiency.
Robust Method for Maximum Efficiency
The covalently bound final product allows use of chemically or electrocompetent cells for optimal transformation efficiency. This increases the likelihood of obtaining correct clones, even for complex constructs. No restriction enzymes, ligation, or recombination sites are required, resulting in a perfectly seamless construct without unwanted extra bases.
