Target Information
Lipolytic enzymes are essential for mobilizing fatty acids from triglyceride stores in adipose tissue. Dysfunctional lipolysis affects energy homeostasis and may contribute to obesity and insulin resistance.
Previously, hormone-sensitive lipase (HSL) was recognized as the main triglyceride-hydrolyzing enzyme in mammalian adipose tissue. However, adipose triglyceride lipase (ATGL) has been identified as another key enzyme catalyzing the initial step in triglyceride hydrolysis.
ATGL is highly expressed in adipose tissue of both mice and humans, exhibits high substrate specificity for triacylglycerol, and associates with lipid droplets. Inhibition of ATGL markedly decreases total adipose acyl-hydrolase activity, indicating that ATGL and HSL work together to catabolize stored triglycerides.
