Target Information
Leukemia inhibitory factor (LIF) is a 20 kDa protein that belongs to the IL-6 receptor family. It binds to a heterodimeric membrane receptor composed of a LIF-specific subunit (gp190 or LIFR) and the shared subunit gp130. LIF is expressed in various tissues such as thymus, lung, and neuronal tissue. Its expression can be up-regulated by pro-inflammatory cytokines like TNFα and IL-17. Elevated LIF levels have been found in rheumatoid arthritis, neural injury, systemic inflammation, and tuberculosis.
LIF is known for its ability to inhibit differentiation of embryonic stem cells in mice, contributing to stem cell self-renewal. Human and mouse LIF share 79% sequence homology and exhibit cross-species activity, although stem cell differentiation inhibition appears to be mouse-specific.
