Target Information

IRF3 (Interferon Regulatory Factor 3) is a key transcriptional regulator of type I interferon (IFN)-dependent immune responses. It plays a critical role in the innate immune response against DNA and RNA viruses by regulating the transcription of type I IFN genes (IFN-α and IFN-β) and IFN-stimulated genes (ISG) through binding to interferon-stimulated response elements (ISRE).

IRF3 is inactive in the cytoplasm of uninfected cells and becomes activated upon phosphorylation by IKBKE and TBK1 kinases following viral infection or toll-like receptor signaling. This activation induces dimerization and nuclear localization, forming the dsRNA-activated factor 1 (DRAF1) complex that activates transcription of type I IFN and ISG genes.

Mutations in IRF3 can lead to acute, infection-induced, herpes-specific encephalopathy.
