Target Information
The PKC family of serine/threonine kinases, including PRKCI (PKC iota), is activated intracellularly by signal transduction pathways. In humans, at least 12 different PKC polypeptides have been identified. These isoforms differ in structure, tissue distribution, localization, and substrate specificity. PKC alpha, beta I, beta II, and gamma form the conventional family, which are Ca²⁺- and phospholipid-dependent.
PKC lambda/iota is calcium-independent and phospholipid-dependent, not activated by phorbol esters or diacylglycerol. It interacts with RAB2 in vesicle tubular clusters (VTCs) to phosphorylate GAPDH, playing a role in microtubule dynamics. This kinase is necessary for BCL-ABL-mediated resistance to drug-induced apoptosis, protecting leukemia cells from apoptosis. A single exon pseudogene is mapped on chromosome X.
