Product Specific Information

The O323 monoclonal antibody reacts with human CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by a subset of thymocytes and virtually all mature T cells and is upregulated upon T-cell stimulation. CD27 binds to CD70 and plays an important role in T cell-B cell interaction.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of normal human peripheral blood cells.
Recommended usage ≤ 5 µL (0.8 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal results.

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.

• Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

NovaFluor conjugates utilize Phiton™ technology with engineered nucleic acid dye structures for optimized fluorescence performance and reduced spillover.

Target Information

CD27 is a 50 kDa member of the TNF receptor superfamily. It regulates cell proliferation and death and is primarily expressed on lymphocytes. CD27 forms disulfide-linked homodimers on mature thymocytes, peripheral blood T cells, and certain B cells. Activation via TCR-CD3 upregulates CD27 and leads to release of soluble CD27 (sCD27), a prognostic marker for B cell malignancies. CD27 dysfunction is associated with Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
