Background

The protein encoded by this gene is a cytokine belonging to the tumor necrosis factor (TNF) ligand family. It acts as a ligand for the FN14/TWEAKR receptor and has overlapping signaling functions with TNF, but exhibits a broader tissue distribution. This cytokine exists in both membrane-bound and secreted forms and can induce apoptosis via multiple pathways in a cell type-specific manner. It also promotes proliferation and migration of endothelial cells, functioning as a regulator of angiogenesis. Alternative splicing produces multiple transcript variants. Some transcripts skip the last exon and continue into the second exon of the neighboring TNFSF13 gene, forming read-through transcripts (GeneID 407977, TNFSF12-TNFSF13). [RefSeq, Oct 2010]
