This gene encodes one of the catalytic subunits of protein kinase A (PKA), which exists as a tetrameric holoenzyme with two regulatory and two catalytic subunits in its inactive form.
Upon cAMP binding, the holoenzyme dissociates into regulatory dimers and free catalytic monomers.
Four regulatory and three catalytic subunits are identified in humans.
PKA-mediated phosphorylation regulates differentiation, proliferation, and apoptosis.
Constitutive activation due to somatic mutations or gene duplication is linked to adrenal cortex hyperplasia and corticotropin-independent Cushing’s syndrome.
