Background

• Function: Transcriptional activator binding to the E-box 1 core sequence (5’CAGGTG-3’) of the E-cadherin promoter gene; capable of reversing CTBP1-mediated transcription repression. Also participates in regulation of alternative pre-mRNA splicing and associates with spliced mRNA near 5’-splice sites. Involved in epithelial cell-cell adhesion and may act as a tumor suppressor in renal cell carcinoma.
• Post-translational Modification (PTM): Phosphorylated upon DNA damage, likely by ATM or ATR.
• Similarity: Belongs to the pinin family.
• Subcellular Location: Cell-cell contact area, predominantly desmosome of intercellular adherens junction. Not a nucleocytoplasmic shuttling protein.
• Subunit: Found in complexes with SR proteins and mRNP complexes with RNPS1. Interacts with multiple proteins including CTBP1, CTBP2, KRT8, KRT18, KRT19, PPIG, RNPS1, SFRS4, SRRM2, and others. Identified in spliceosome C complex.
• Tissue Specificity: Expressed in placenta, lung, liver, kidney, pancreas, spleen, thymus, prostate, testis, ovary, small intestine, colon, heart, epidermis, esophagus, brain, and muscle. Strongly expressed in melanoma metastasis lesions and advanced primary tumors.
