Background
The PH domain of PHLDA3 binds phosphoinositides with broad specificity and competes with the PH domain of AKT1, interfering with AKT1 binding to PIP2 and PIP3. This prevents AKT1 association with membrane lipids and subsequent activation of AKT1 signaling.
PHLDA3 is a p53/TP53-regulated repressor of AKT1 signaling, contributing to p53-dependent apoptosis by repressing AKT1 activity. Expression is directly activated by p53/TP53, and phosphorylation on Ser-15 is required for PHLDA3 promoter activation.
The PHLDA3 genomic locus is frequently altered in primary lung cancers, suggesting a tumor suppressor role. It belongs to the PHLDA3 family and contains one PH domain. Expression is widespread, with the lowest levels in liver and spleen.
