Background
This gene encodes a potent cyclin-dependent kinase inhibitor. The encoded protein binds to and inhibits the activity of cyclin-CDK2 or cyclin-CDK4 complexes, regulating cell cycle progression at G1. Its expression is controlled by the tumor suppressor p53, mediating p53-dependent G1 arrest under stress. The protein interacts with proliferating cell nuclear antigen (PCNA) and plays roles in DNA replication and repair. It can be cleaved by CASP3-like caspases, activating CDK2 and contributing to apoptosis execution.
