Background
This gene lies adjacent to the tumor suppressor gene CDKN2A in a region frequently mutated and deleted in various tumors. It encodes a cyclin-dependent kinase inhibitor that forms a complex with CDK4 or CDK6, preventing activation of these kinases and thereby regulating cell cycle G1 progression. Expression of this gene is dramatically induced by TGF-β, suggesting its role in TGF-β–induced growth inhibition. Two alternatively spliced transcript variants encoding distinct proteins have been reported.
