Target Information
BRCA1-associated protein-1 (BAP1)은 BRCA1의 RING finger domain과 상호작용하는 단백질입니다. 인간 BAP1 단백질의 N-말단 240 아미노산은 ubiquitin C-terminal hydrolases (UCHs)와 상동성을 가지며, 이는 ubiquitin의 단백질 분해 가공을 촉매하는 효소입니다.
BAP1은 산성 영역, 고전하 C-말단 영역, 두 개의 핵 내 위치 신호를 포함하고 있으며, BRCA1과 세포핵 내에서 공존하며 유방암 세포 성장 억제를 강화합니다.
Northern blot 분석에 따르면, BAP1은 대부분의 인간 조직에서 4-kb mRNA로 발현되며, 4.8-kb 전사체는 고환에서만 특이적으로 발현됩니다. 또한 BAP1과 BRCA1은 생쥐 유방 발달 및 재형성 과정에서 함께 발현됩니다.
BAP1 유전자는 3p21.3에 위치하며, 이 영역은 유방암 및 폐암에서 종종 결실되는 영역으로 알려져 있습니다. BAP1은 BRCA1 성장 조절 경로에서 종양 억제 유전자로 작용할 가능성이 제시되었습니다.
