Product Specific Information
- For dilution, use a buffer containing carrier protein (e.g., PBS with 1% BSA) or neutral media containing 10% FCS.
- Freeze aliquots at -70°C.
- Activity: Biological activity measured by cytolytic effect on mouse WEHI-13VAR cells in presence of Actinomycin D.
- Expression: Mature, soluble form of equine TNF-alpha expressed in Pichia pastoris KM71H (GenBank® Accession # P29553, amino acids 78–234).
- Reconstitute with 0.5 mL of distilled water.
Target Information
TNF-alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR.
This cytokine regulates immune cells, proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It exists as multimers but appears as a single 17 kDa band under non-reducing SDS-PAGE conditions.
Knockout studies suggest TNF-alpha has neuroprotective functions and can induce tumor necrosis in mice. It plays roles in immune responses to bacterial, viral, parasitic, and fungal infections and contributes to the necrosis of specific tumors.
TNF-alpha causes cytolysis or cytostasis of certain transformed cells, synergistic with interferon-gamma. It is implicated in autoimmune diseases, insulin resistance, and cancer.
