The LIVE/DEAD™ Fixable Violet Dead Cell Stain Kit is designed to determine cell viability prior to fixation and permeabilization for intracellular antibody staining or before the elimination of biohazardous materials using formaldehyde fixation. This kit is optimized and validated for use with violet laser flow cytometers.

Stability

Unlike solution-based products, this stain is provided in 40-test vials to ensure dye stability and performance. Once rehydrated, use the vial completely or aliquot into small volumes and store at -80°C, avoiding freeze-thaw cycles.

Robustness

The amine-reactive dye binds covalently to intracellular and extracellular amines, preserving staining patterns after formaldehyde fixation. It maintains sensitivity even after fixation methods used in intracellular phosphorylation studies.

Low Compensation

This violet-fluorescent dye minimizes compensation between other violet and blue laser dyes.

• Excitation maximum: ~416 nm (optimized for 405 nm laser)
• Emission maximum: ~451 nm (collectable in first violet channel)

How It Works

In cells with compromised membranes, the dye reacts with free amines inside and on the surface, yielding intense fluorescence. In viable cells, reactivity is limited to surface amines, resulting in weaker fluorescence—typically a >50-fold difference between live and dead cells.
