• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

Cyclin E belongs to the highly conserved cyclin family, characterized by periodic protein abundance during the cell cycle. Cyclins regulate CDK kinases and exhibit distinct expression and degradation patterns that coordinate mitotic events.
Cyclin E forms a complex with CDK2, regulating the G1/S transition. It accumulates at the G1-S boundary and degrades during S phase. Overexpression is observed in many tumors, contributing to chromosome instability and tumorigenesis.
Cyclin E associates with and phosphorylates NPAT, which regulates histone gene expression and promotes cell-cycle progression in the absence of pRB. Two alternatively spliced transcript variants encode distinct isoforms; additional variants have been reported but are not fully characterized.
