Target Information

Phosphorylase kinase is a hexadecameric enzyme composed of four copies of four subunits encoded by PHKA, PHKB, PHKG, and PHKD genes. This serine/threonine-specific kinase converts glycogen phosphorylase b to glycogen phosphorylase a, releasing glucose-1-phosphate from glycogen.

PHKB (Phosphorylase b kinase regulatory subunit beta) is a 1093 amino acid regulatory subunit of phosphorylase kinase. Along with PHKA, it plays a regulatory role controlled by phosphorylation. Mutations in the PHKB gene cause glycogen storage disease type 9B (phosphorylase kinase deficiency of liver and muscle), characterized by mild hepatomegaly, slightly elevated transaminase and plasma lipids, and general improvement with age. Four isoforms of PHKB exist due to alternative splicing.
