Target Information
KIT (c-KIT)은 MGF(mast cell growth factor, stem cell factor)의 수용체로 작용하는 type 3 transmembrane receptor이자 proto-oncogene입니다. KIT은 처음에 feline sarcoma viral oncogene v-kit의 세포 상동체로 확인되었습니다. KIT과 그 리간드는 다양한 조혈 및 비조혈 세포의 성장과 활성화를 조절합니다. KIT의 변이는 위장관 기질종양(GIST), 비만세포 질환, 급성골수성백혈병 및 piebaldism 등과 관련이 있습니다. 특히 D816V 활성 루프 변이에 의한 KIT 수용체의 탈조절은 비정상적인 비만세포 증식을 억제하기 위한 치료 전략의 주요 표적이 되고 있습니다.
c-Kit은 조혈 줄기세포, 생식세포, 비만세포, 위장관의 Cajal 세포에서 발현됩니다. SCF(stem cell factor)가 결합하면 c-Kit은 이합체화되어 수용체가 활성화되고, 세포질 도메인 내의 여러 티로신 잔기(예: Tyr703)가 자가 인산화됩니다. 이 과정은 Grb2의 도킹과 Ras/ERK 신호경로의 활성화를 유도합니다. 또한 SCF/c-Kit은 PI3K, PLC-γ, JAK/STAT 등 다양한 하위 신호경로를 활성화할 수 있습니다. c-Kit 수용체의 활성화는 조혈, 줄기세포 유지 및 생식세포 형성에 필수적입니다.
