Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. Upon stress, p53 activates transcription of specific genes such as p21waf1, regulating G1 or G2/M arrest. Structurally, p53 consists of an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.
Phosphorylation at Ser15 is critical for p53 activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 also plays roles in aging, oxidative stress, and cognitive functions, with elevated levels linked to deficits in long-term potentiation (LTP). Normally expressed at low levels, p53 is upregulated in transformed cells, contributing to malignancy. Mutations in p53 disrupt DNA binding and tumor suppression, and alterations in the TP53 gene are found in both somatic and germline mutations, including Li-Fraumeni syndrome.
