Target Information
Tight junctions form an important barrier for paracellular transport in epithelial cells. Tricellulin is the first protein identified that specifically concentrates in tricellular tight junctions — points where three adjacent cells meet. This protein has four membrane-spanning domains, similar to claudins. Tricellulin is highly expressed in epithelium-derived tissues such as the small intestine, kidney, and lung. Functional studies using siRNA show that suppression of tricellulin expression compromises epithelial barrier integrity and tight junction formation. Mutations affecting the cytosolic domain, which binds to the scaffolding protein ZO-1, suggest that interactions with other tight junction proteins are critical for tricellulin function.
