Caspases are present in all cells, and recruitment of these proteases to oligomerized receptors leads to activation accompanied by autoproteolytic cleavage. Active caspases can proteolyze additional caspases, generating a cascade that cleaves proteins critical for cell survival. The final outcome of this signaling pathway is controlled cell death, termed apoptosis.
Caspase-3 cleaves substrates at the carboxyl terminus of aspartate residues. Active caspase-3 consists of two identical large (~20 kDa) and two identical small (~10 kDa) subunits derived from precursor polypeptides. Both subunits contribute to substrate binding and catalysis, with the active site cysteine near the C-terminus of the large subunit. Caspase-3, together with caspases 8 and 9, plays a pivotal role in apoptotic pathways, amplifying initiation signals into full apoptotic disassembly.

Target Information
This gene encodes a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution phase of apoptosis. Caspases exist as inactive proenzymes that undergo proteolytic processing to form active enzymes composed of large and small subunits. Caspase-3 cleaves and activates caspases 6, 7, and 9, and is itself processed by caspases 8, 9, and 10. It is a predominant caspase involved in cleavage of amyloid-beta precursor protein, associated with neuronal death in Alzheimer’s disease. Alternative splicing results in two transcript variants encoding the same protein.
