Target Information
ZBRK1 (ZNF350) is a member of the Kruppel-associated box-zinc finger protein (KRAB-ZFP) family of transcriptional repressors. It is up-regulated in senescent fibroblasts and interacts with BRCA1, a transcriptional regulator with tumor suppressor activity.
ZBRK1 binds to specific sequences within the GADD45 gene, forming a nuclear complex with BRCA1 to repress transcription. It can homo-oligomerize through a C-terminal transcriptional repression domain (CTRD), facilitating transcriptional repression via ZBRK1 response elements.
These findings suggest that higher-order ZBRK1 oligomers may assemble through both protein-DNA and CTRD-dependent protein-protein interactions.
