Target Information
The p38 MAPK cascade regulates various cellular responses to stress, inflammation, and other signals. p38 MAPK is relatively inactive in its nonphosphorylated form and becomes rapidly activated by dual phosphorylation of a Thr-Gly-Tyr motif. There are four isoforms of p38 MAPK, differing in tissue expression and affinity for upstream activators and downstream effectors. Upon exposure to tumor necrosis factor, interleukin-1, heat shock, or other stimuli, MAPK kinase-3 becomes phosphorylated and activates MAPK kinase-3/6, which phosphorylate Thr180 and Tyr182 residues in p38 MAPK. Activated phospho-p38 MAPK subsequently phosphorylates transcription factors such as ATF-2, CHOP-1, and MEF-2.
