Product Specific Information
This 4LURPIE monoclonal antibody recognizes human and mouse STAT4 when phosphorylated on tyrosine 693 (Y693). STAT proteins are activated by cytokine receptor binding and subsequent phosphorylation by JAK kinases, leading to nuclear translocation and transcriptional regulation.
STAT4 is activated by IL-12 or type I interferons (IFN-α, IFN-β) and is essential for Th1 differentiation. In mice, IFN-α induces transient and weak STAT4 phosphorylation compared to IL-12, thus contributing minimally to Th1 development, whereas in humans, IFN-α promotes Th1 differentiation and IFN-γ production.
Specificity of clone 4LURPIE was confirmed by ELISA and flow cytometry.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated for intracellular staining of stimulated human peripheral blood cells at 5 µL (0.25 µg)/test.
A test is defined as the amount of antibody used to stain cells in a 100 µL final volume (typically 10⁵–10⁸ cells/test).
Recommended Staining Protocol:
Use Protocol C (Two-step: Fixation/Methanol).
Protocols A (cytoplasmic proteins) and B (nuclear proteins) are not suitable.

Target Information
STAT4 shares sequence similarity with STAT1 (52%) and STAT3 (47%) and can be phosphorylated by Jak1 or Jak2. Phosphorylated STAT4 forms homo- and heterodimers, binds IFN-γ activated sites (GAS), and requires serine phosphorylation for maximal activity.
STAT4 expression is mainly in thymus, spleen, and testis. It is activated by IL-12, which drives IFN-γ induction and Th1 differentiation crucial for defense against intracellular pathogens. STAT4-deficient mice show impaired Th1 development and enhanced Th2 differentiation. During viral infection, IFN-α/β activation of STAT4 is essential for IFN-γ production.
