Target Information
TNK2 (ACK1), also known as activated Cdc42-associated kinase, is a non-receptor tyrosine kinase that functions downstream of Cdc42. It is implicated in tumorigenesis and metastasis, especially in prostate cancer. TNK2 mediates CDC42-dependent cell migration via phosphorylation of BCAR1 and may play roles in synaptic function, brain development, and regulation of multiple signaling pathways.
Key functions include:
- Activation of AKT1 via phosphorylation at Tyr-176
- Phosphorylation of AR at Tyr-267 and Tyr-363, enhancing recruitment to androgen-responsive enhancers
- Regulation of WWOX, MCF2, and AXL receptor pathways
- Promotion of metastatic potential and tumor growth through modulation of pro-survival and tumor-suppressor factors
