Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family that plays an integral role in T cell signal transduction, differentiation, and activation.
CD8 is primarily expressed on T cells as a disulfide-linked heterodimer (CD8α and CD8β), functioning as a co-receptor with the T cell receptor (TCR) for MHC class I molecules.
In contrast, CD4 acts as a co-receptor for MHC-II molecules. CD8α is essential for MHC-I binding and is also expressed on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers.
Engagement of MHC-I/peptide complexes recruits Lck kinase, leading to lymphokine production and cytotoxic T lymphocyte (CTL) activation, which are key to pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
