This product is diluted and ready-to-use.
A recommended positive control tissue is Ewing’s sarcoma, though other positive controls may be used.
The antibody is intended for professional laboratory use in detecting NKX2.2 protein in formalin-fixed, paraffin-embedded tissue through manual qualitative immunohistochemistry (IHC). It is used after the primary diagnosis of tumor by conventional histopathology.
NKX2.2 expression is observed in neuroendocrine tumors of the gut and serves as a marker for gastrointestinal neuroendocrine tumors. It is also a target of the EWS-FLI-1 fusion protein specific to Ewing sarcoma, showing 93% sensitivity and 89% specificity, aiding in differential diagnosis of small round cell tumors.
Pretreatment of deparaffinized tissue with heat-induced or enzymatic epitope retrieval is recommended.
IHC staining involves sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate, with washing steps between applications. The chromogen reaction produces visible staining at the antigen site, interpreted under a light microscope.
A positive tissue control must be included in each staining procedure. This control should contain both positive and negative staining regions and be processed identically to patient samples. Weak positive controls are preferred for detecting minor changes in antibody sensitivity or reagent stability.
Negative control tissue may be internal or external depending on institutional guidelines. Negative sites should show absence of specific staining, ensuring minimal background signal. If specific staining appears in negative controls, patient results are invalid.
Target Information
The NKX2.2 gene encodes a nuclear transcription factor containing a homeobox domain, involved in central nervous system morphogenesis. It is located on chromosome 20 near NKX2-4 and duplicated on chromosome 14 as TITF1 and NKX2-8.
