Target Information
The family of voltage-dependent chloride channels (CLCs) regulate cellular trafficking of chloride ions, which are essential for cellular excitability, solute transport, and volume maintenance.
- CLCN1 is highly expressed in skeletal muscle. Mutations in CLCN1 cause myotonia, characterized by muscle stiffness and renal salt wasting.
- CLCN2 is expressed in epithelial tissues such as lung and may be a therapeutic target for cystic fibrosis.
- CLCN3 is abundant in neuronal tissue.
- CLCN4 is found in skeletal, cardiac muscle, and brain.
- CLCN5 mutations cause Dent’s disease (proteinuria, hypercalciuria).
- CLCN6 and CLCN7 are broadly expressed in testis, kidney, brain, and muscle.
