Product Specific Information

The GK1.5 monoclonal antibody reacts with mouse CD4, a 55 kDa cell surface receptor expressed on most thymocytes, subsets of mature T cells, and dendritic cells. CD4 binds to MHC class II molecules on antigen-presenting cells, playing a critical role in T cell development and activation. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Applications Tested: Tested on mouse thymocytes and splenocytes at ≤0.25 µg/test.
A test is defined as the amount of antibody staining a sample in 100 µL final volume. Cell count may range from 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen™ LIVE/DEAD™ Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails before mixing with cells.
  • For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescence with minimal spillover and spread.

Target Information

CD4 is a co-receptor for MHC class II molecules and plays a critical role in T-cell activation. It is mainly expressed on T helper cells, with additional expression in monocytes, macrophages, and dendritic cells. CD4 is also found in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues (e.g., brain, gut). It associates with the T-cell receptor complex and Lck kinase, contributing to immune signaling and, in some cases, neuronal damage in immune-mediated diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
